Bacterial reverse mutation test (Ames test)

Aim:

The purpose of this study is to evaluate the mutagenic potential of the test item by measuring its ability to induce reverse mutations at selected loci of several strains of Salmonella typhimurium and at the tryptophan locus of Escherichia coli strain WP2 uvrA in the presence and absence of metabolic activation by rat liver S9.

Test system:

Salmonella typhimurium and Escherichia coli strains

Design:

Applied concentrations are determined in the Concentration Range Finding Test (Informatory Toxicity Test) and Solubility Tests.

The results of the different concentrations are compared to untreated, vehicle treated and positive controls (all with presence and absence of metabolic activation by rat liver S9).

Outcome:

The test item is considered mutagenic, if

  • a dose-related increase in the number of revertants occurs and
  • a reproducible biologically relevant positive response for at least one of the dose groups occurs.
