The aim of this type of study is to assess the delayed-type of hypersensitivity reactions and identify skin-sensitizing substances.
The basic principle of this test is that potential sensitizing test items induce proliferation of lymphocytes in the lymph nodes draining the site of application. This proliferation is proportional to the dose and to the potency of the applied allergen.
At least three dose groups, one vehicle control and one positive control group are used. The diluted test item is applied on multiple days on each ear of the mice and the draining auricular lymph node is analyzed.
The proliferation in the lymph nodes in the treated groups is compared to the proliferation in the control animals, and the Stimulation Index (ratio of the mean proliferation in the treated group to that in the control group) is determined.
